PureStem™ E15, Meso-prx/latp Progenitor

The clonal human embryonic progenitor line E15 expresses chondrogenic markers (SOX9, VCAN, COL6A2, COL1A1) and markers of axial and appendicular derived mesenchymal cells (TFAP2A, CDH2, SIX1). 

Progenitor line E15 was derived from the parental NIH registered Human ES cell line ACT03 (MA03) (West, M.D. et al, 2008) 1.

Differentially expressed positive mRNA markers (by microarray):

VCAN, TMEM132D, TFAP2A, TBX15, SOX9, SIX1, PODXL, MAP3K8, KAL1, COL6A2, CDH2, AJAP1

Selected cell surface markers:

CDH2, CDH6, EPHA5

Selected secreted factors:

COL1A1, COL1A2, COL6A2, FBLN2, GDF5, HAPLN1, NGF, POSTN, VCAN

Experimental Data

  • PureStem E15 was characterized by MicroArray analysis and compared to bone marrow-derived Mesenchymal Stem Cells - see heat map below.
  • PureStem E15 were induced to differentiate in vitro into functional chondrocytes.

Growth Media:

Optimal growth conditions and growth factors are detailed in the product datasheet.
LifeMap recommends using progenitor growth media em 1001, which is optimized for growth of E15 mesenchymal progenitor cells.

Differentiation Protocols:

The Chondrogenic differentiation protocol is detailed in datasheet (purestem chondrogenic differentiation kit em 2001).

Propagation Potential:

PureStem Mesenchymal Progenitor E15 is sold at passage 13 (passage 1: original clonal isolate at confluence in 1.9 cm2 well), which corresponds to approximately 16.5 doublings since the first passage and approximately 33 doublings since its original clonal plating. The line displays a finite lifespan in vitro, and when properly maintained may reach 80 population doublings.

Quality Control:

The line is negative for HIV (1, 2), HBV, HCV, bacteria, mycoplasma, and fungal contamination. Certificates of analysis are available on request.

About HyStem Hydrogel

The HyStem Hydrogel is a xeno-free Hyaluronan-based hydrogel product designed for optimal culturing of cells. HyStem can be customized by adding extracellular matrix (ECM) proteins or cell attachment peptides into the hydrogel to provide attachment site and/or differentiation signals. HyStem Hydrogels can also be varied by changing the hydrogel rigidity to match that of the native cell environment.
