The clonal human embryonic progenitor line SK11 expresses chondrogenic (COL1A1, VCAN, SOX9, COL6A1) and osteogenic (RUNX2, ALPL, SPP1) markers. In addition, it expresses markers of axial and appendicular level-derived mesenchymal cells (SIX1, TWIST1, SNAI2). The cell line exhibits chondrogenic and osteogenic fates upon differentiation in vitro.

Progenitor line SK11 was derived from the parental NIH registered Human ES cell line H9 (WA09) (West, M.D. et al, 2008) 1.

Experimental Data

Heat map of unique gene expression markers distinguishing Mesenchymal stem cells (MSCs) and SK11 in the undifferentiated state. The relative expression of differentially-expressed marker genes detected by Illumina microarray analysis by comparing RNA from MSCs and SK11.

  • PureStem SK11 was characterized by MicroArray analysis and compared to bone marrow-derived Mesenchymal Stem Cells - see heat map below.

Growth Media:

LifeMap recommends using progenitor growth media em 1005, which is optimized for growth of SK11 mesenchymal progenitor cells.

Differentiation Protocols:

Chondrogenic and Osteogenic differentiation protocols are detailed in their respective differentiation kits' datasheets (purestem chondrogenic differentiation kit em 2001 and osteogenic differentiation kit em 2003).

Propagation Potential:

PureStem Mesenchymal Progenitor SK11 is sold at passage 11 (passage 1: original clonal isolate at confluence in 1.9 cm2 well), which corresponds to approximately 13.5 doublings since the first passage and approximately 30 doublings since its original clonal plating. The line displays a finite lifespan in vitro, and when properly maintained may reach 70 population doublings.

Quality Control:

The line is negative for HIV (1, 2), HBV, HCV, bacteria, mycoplasma, and fungal contamination. Certificates of analysis are available on request.

About HyStem Hydrogel

The HyStem Hydrogel is a xeno-free Hyaluronan-based hydrogel product designed for optimal culturing of cells. HyStem can be customized by adding extracellular matrix (ECM) proteins or cell attachment peptides into the hydrogel to provide attachment site and/or differentiation signals. HyStem Hydrogels can also be varied by changing the hydrogel rigidity to match that of the native cell environment.
